Protein Function
Translocates drugs and phospholipids across the membrane (PubMed:8898203, PubMed:2897240, PubMed:9038218). Catalyzes the flop of phospholipids from the cytoplasmic to the exoplasmic leaflet of the apical membrane. Participates mainly to the flop of phosphatidylcholine, phosphatidylethanolamine, beta-D-glucosylceramides and sphingomyelins (PubMed:8898203). Energy-dependent efflux pump responsible for decreased drug accumulation in multidrug-resistant cells (PubMed:2897240, PubMed:9038218).

  • Decreased clearance of Ripretinib due to the transporter inhibition by Simeprevir 

Recommended Action
      Management Until more information is available, caution is recommended when ripretinib is used concomitantly with drugs that are inhibitors of P-gp. Clinical and laboratory monitoring should be considered whenever a P-gp inhibitor is added to or withdrawn from therapy with ripretinib, and the dosage adjusted as necessary. Patients should be monitored for development of adverse effects such as nausea, vomiting, constipation, diarrhea, alopecia, arthralgia, myalgia, hypertension, cardiac dysfunction, and palmar-plantar erythrodysesthesia.
